Rapid Total RNA Purification Systems are designed for the fast isolation
of total RNA from a wide variety of cells and tissues. Based on differential
binding of total RNA to silica membranes, these products are optimized
for the separation of RNA from DNA, proteins and other cellular components.
They are compatible with either vacuum or centrifugation protocols and
are available in both individual column and 96 well plate formats. The
Rapid 96™ Systems provide fast and reliable purification of total
RNA in a 96 well plate format. The Rapid Total RNA System (column format)
can be used for both mini and midi RNA purification applications. RNA
isolated with the Rapid Total RNA Purification System is suitable for
direct analysis in a variety of applications or as a substrate for the
preparation of mRNA.
Technology Marligen’s Total RNA Isolation Kits rely on disrupting cells and tissues in the presence of chaotropes, which also simultaneously inactivate endogenous nucleases, thereby protecting the RNA from degradation. The solubilized RNA is then adsorbed to a silica membrane under conditions that optimize binding of RNA versus contaminants. Following washes to remove remaining contaminants, the RNA is eluted, ready to use in a variety of applications including RT-PCR, microarray analysis, Northern blotting, RNase protection assays or cDNA library construction. Performance Characteristics Figure 1 shows that RNA isolated with Marligen’s Rapid RNA Isolation Kits demonstrates high yields of intact RNA. Figure 2 shows that A260/280 ratios, total yield and overall concentration is very reproducible from prep to prep. Each lot of kits is tested by agarose gel electrophoresis and spectrophotometric analysis to verify the purity and integrity of the resulting RNA.
Fig. 1 Three samples containing 107 Hela cells were purified with the Marligen Total RNA Purification System. Gel results demonstrate a high yield of intact RNA and high reproducibility.
Table 1 below shows data from the purification of total RNA from three identical samples of 107 Hela cells shows excellent purity and yield. RNA is eluted at a high concentration and reproducibility is high with coefficients of variation below 10%.
